Detection and determining distribution of cucumber mosaic virus in fields located in Khorasan province

Investigations were conducted during 2001 to detect and determining distribution of cucumber mosaic virus (CMV) in fields located in northern parts of Khorasan province. Leaves showing chlorosis, mosaic, distortion and shoestring symptoms were collected and transferred to laboratory for CMV identification and further studies. To investigate the distribution of CMV in Mashhad, Fariman, Neishabour, Chenaran, Ghochan, Shirvan and Torbat-e-heidarye regions, samples were collected randomly. Out of 500 samples tested by DAS-ELISA, 208 were infected with CMV. This survey showed that infection rate was higher in Chenaran and Torbat-e-heidarye compared to other regions were tested in this project. Samples collected from Torbat-e-heidarye were used for inoculating tobacco plants grown under greenhouse condition. These plants were also used in RT-PCR test and a 637bp fragment of DNA corresponding to virus genomic RNA2 was amplified in samples infected with CMV.
