Study of the potential of DAS-ELISA technique in optimization of chemical control against wheat leaf blotch disease caused by Septoria tritici

Forecasting of occurrence of septoria tritici leaf blotch is very important for precise adjustment and designing of chemical control programs. One of the methods of pre-symptomatic identification of this disease is the use of immunoassays techniques. In this research. the importance of application of this technique was studying for determining of fungicide mode and dosage and spray timing. For this purpose, Roshan wheat cultivar was sown in 44 pots. After twelve days, seedlings were inoculated with sporidial suspension. The experiment was conducted using a completely randomized block design as factorial with three replications. The first 20 pots pre-symptomaticaly and second 20 pots postsymtomaticaly were treated by 2, 1 and 0.5 per thousands of Tilt and Alto fungicides. 1 he rest 4 pots were treated with tap water for controls. After providing of leaf extract and conduction FLISA test, the optical densities (O.D.) of samples were determined and convert in Antigenic unites (AGU) and the effect of treatment on AGU was compared with one another. Results indicated there is significantly difference between pre and post-symptomatic sprayings. We can decrease spraying dose and thus control cost with forecasting of this disease by the use of these detection kits. It wasnʹt observed significant effect between decreasing percent in the use of Tilt and Alto. Besides determining of accurate timing for spraying, application of this technique will he resulted in decreasing of dosage and more effective fungicides.
