The Effect of Sodium Valporate and Folic Acid on The Weight and Development of Limb BUD in Mouse Embryo

Introduction: Valproic acid, a Fatty acid, is usually administered as a sodium valporate. Valproic acid has become accepted as one of the most important antiepileptic drugs. It is teratogenic in humans, mice, rats and monkeys. The mechanism of the teratogenicity of VPA is unknown. One hypothesis is that VPA interacts with embryonic folate metabolism. Materials and Methods: In this study the effectiveness of sodium valporate and folic acid on the weight and development of limb bud was assessed in two experimental groups and one control group. In the first two groups of animals, 200 mg/kg of VPA and in the second group 200 mg/kg of VPA and 4 mg/kg folic acid was injected intraperitaneally on the 8th gestation day. Thirteen days after gestation, the fetus was removed from the uterus and was examined macroscopically and microscopically. Results: This study revealed that VPA reduces the weight of embryos, and induces limb malformation as out pocketings from the ventrolateral body wall. The mean weight of the Control group, group I and group II of animals were 0.086 gr, 0.021 gr and 0.0619 respectively. The P value between the Control group and group I, and also between group I and group II was less than 0/05. Folic acid can prevent weight loss and abnormalities in limb bud of embryos. Conclusion: VPA decreases the weight of fetuses and lead to limb malformation as it reduces the supply of nutrients to the embryo and the proliferation of the vessels. VPA mutation in 10.5 methylene tetra hydrofolate reductase results in limb malformation and weight loss.
