A Study of Seed Dormancy, after-Ripening and Polypeptide Patterns of Dormant and Non-Dormant Embryos in Bread Wheat (Triticum aestivum)

To study seed dormancy and after-ripening, 30 bread wheat cultivars were sown in a randomized complete block design with three replications at the Research Field of Agriculture Faculty of the University of Tehran in 2002. Days to 50% spike emergence, days to 50% anthesis, and Zadoksʹ growth stage 92 (ZGS92) were recorded. At ZGS92 spikes were harvested and the grain moisture content measured for all cultivars. Seed dormancy tests were conducted at two temperatures (10°c, 20°c) and seed dormancy index calculated. To determine length of seed dormancy period, after-ripening tests were conducted for six weeks. For Alphaamylase activity, Falling number test was used. Results indicated that there were significant genetic variations among Iranian bread wheat cultivars for all aforementioned traits. These variations can be used in breeding programs for pre-harvest sprouting resistance in bread wheat. To test the hypothesis that differential protein synthesis is responsible for maintaining seed dormancy in wheat, we the polypeptide patterns of dormant and non-dormant embryos in wheats were compared. The results demonstrate that steady-state levels of specific proteins differ between dormant and non-dormant embryos during early imbibitions, and these differences may be associated with maintaining andlor breaking seed dormancy.
