Immuno double diffusion test and indirect enzyme linked immunosorbent assay (indirect ELISA) were evaluated for identification and detection of Xanthomonas axonopodis pv. phaseoli (Xap) and X axonopodis pv. phaseoli var. fuscan (Xapf) in bean seed. Antisera were prepared against fonnalin- fixed and live bacterial cells of Xap- 14 and Xapf- 3. The Antisera reacted strongly with different isolates of Xap and Xapf in both serological methods. No serological reaction was observed when antisera were tested against unrelated bacteria as well as different strains of Xanthomonas sp. Although double diffusion test was considered as a specific and reliable method for identification of Xap and Xapf, the method was not sensitive enough and at least l0^8 cfu/ ml was required for band formation. The isolates Xap and Xapf were differentiated from each other by using their Xanthomonadin in double diffusion test. In indirect ELISA method, it was possible to detect bacteria at concentration of approximately l05 du/ ml. The use of ELISA for detection of Xap from naturally infected seeds soaked in sterile water showed that, this method could be a suitable and sensitive one for detection of the causal agent of common bacteria blight of bean in seedlots.
