PURIFICATION AND STRUCTURAL CHARACTERIZATION OF THE MAJOR GL YCOCONJUGA TE FROM LEISHMANIA. L.MAJOR (MRHO/IR/75/ER)

Lipophosphoglycan (LPG) is a glycoconjugate present on the surface of leishmania parasites that has been reported to promote intracellular survival of these parasites . It is a lipophosphoglycan containing 81.6% (wt/wt) carbohydrate, 17.0% (wt/wt) phosphate, and 1.4% (wt/wt) lipid. LPG consists of four domains: (a) a phosphatidylinositol lipid anchor, (b) a phosphosaccharide core, (c) a repeating phosphorylated saccharide region and (d) a small oligosaccharide cap structure. We cultured the cloned virulent leishmania . L. major parasite (MRHO/IR/75/ER strain of IRAN). Promastigotes were grown Invitro in NNN meduim (sigma) supplemented with 10% (vol 1 vol) heat-inactivated fetal calf serum(sigma). Promastigotes (5x10^4 cites) were harvested and then preserved for extraction and purification of LPG. In this study , the amount of leishmania.L. major LPG purified was approximately 1 gram.
