Embryo transfer is one of the most important methods for conserving precious genetic reservoirs in agriculturally important species, including cattle. Naturally, 50% of the calves are male or female, so, sex determination before transferring the embryo has a major impact on animal breeding. This study was conducted to determine the sex of domestic and Iranian Holstein cattle, using polymerase chain reaction (PCR) method. Twenty male and 20 female cows (10 domestic and 10 Iranian Holstein for each sex) were randomly selected from different farms in Isfahan. Two pairs of primers, which recognized bovine Y chromosome-specific sequences and are amplified in males only, and a pair of primers which recognized satellite sequences which is amplified in both females and males, were used in PCR. Male specific primers produced DNA fragments of 301 bases, while satellite DNA primers produced DNA fragments of 216 bases. The assay was extremely accurate as no single incorrect result occurred. No differences were observed between Iranian domestic and Holstein tattles.
