In this study, seeds of Medicago sativa c.v. Rehnani, were cultured on MS medium then callus produced from stem segments of invitro grown plants on MS medium containing NAA, 2,4-D and kinetin. Calluses then were transferred on the same media supplemented with 0, 2, 5, 10, 20, 30% PEG (6000 MW). Selection was conducted using direct and step-by-step method. Selected calli for drought tolerance in the stepwise selection showed better growth than direct method under drought stress. However, the selected calli in stepwise and direct methods were able to tolerate drought stress up to 20% and 30% PEG, respectively.
