Aims. In this study, the oxidative stress of diazinone and the vitamin B6 (pyridoxine) protective effect against diazinone in astrocytes type U373MG were evaluated.
Material & Methods. The cells exposed to different doses of diazinone at different time intervals. Then, the effect of 300 pMof diazinone for 72 hours as optimum dose alone or in the presence of vitamin B6, either 30 min before or post diazinone treatment was evaluated on GSH level and cells viability. Results. Diazinone, at the dose of 300 jjM, significantly reduced the GSH level at 72 hours post exposure. Pre-treatment with vitamin B6 could increase the GSH level approximately near the control value. Diazinone could decrease (about 20%) the cells viability, significantly. Either pre- or post-treatment with vitamin B6, could prevent cell death induced by diazinone.
Conclusion. Diazinone probably is toxic for U373MG via oxidative stress induction and decrease of GSH level. Either pre- or post-treatment by vitamin B6 can protect cells and prevents GSH level reduction and cell death.
