2,3-diphosphoglycerate is the most important compound in hemoglobin affinity to oxygen molecules. It reduces the hemoglobin affinity to oxygen molecules and is the most frequent phosphorylated compound in RBC. In this study, a total of 88 blood samples were collected from healthy sheeps according to their and age groups (1-14 day, 14-30 day, 1-2,2-3,3-6, 6-12,12-18, 18-24, 24-36, 36-48 month). The levels of 2, 3-diphosphoglycerate and total hemoglobin were measured by spectrophotometery method and using the Sigma kit (Sigma, Co., 665-PA, Germany) and met-hemoglobin method, respectively. According to the results, there was a significant difference between age groups with 2,3-diphosphoglycerate level (P<0.05), the maximum levels of 2,3-diphosphoglycerate was observed in lto7-day lambs and the minimum levels was in after 2-3 month age groups but within each age groups, there was no a significant difference according to sex concerning the total hemoglobin and 2, 3-diphosphoglycerate levels. In the studied sheeps, there was no significant difference between sex and age groups concerning total hemoglobin, only a negative significant correlation was observed between age groups concerning 2, 3-diphosphoglycerate levels (P<0.05, r = - 0.760).
