Design and Validation of a Competitive ELISA (C-ELISA) for Diagnosis of Brucellosis

Brucellosis has been endemic in Iran, Detection of Brucella antibodies is a useful method for diagnosis. The purpose of this study was to evaluate the performance of the competitive ELISA (CELISA) for the diagnosis of human brucellosis in comparision to routine serological methods. Brucella abortus S99 was used for LPS preparation. The LPS was extracted by hot phenol method with enzymatic digestion. Microtitre plates were coated with LPS. For CELISA antibodies was revealed by using peroxidase conjugated mouse monoclonal antibody M84 for competition. The cutoff values were set at 53.29-56.91% inhibition (1%) for CELISA. The cutoffs were obtained by testing negative serum samples. The specificity for CELISA was 99,5%, and sensitivity was 96.94%,. The positive predictive value for CELISA was 98.96%, and negative predictive value for was 98.51% . Also overall agreement was 98.66%. The results indicate the potential use of the CELISA as a sensitive, specific, rapid and simple methods for diagnosis of human brucellosis.
