A comparative study of productive Yield of Brucella abortus Biomass between Batch and Fed-batch methods

Brucella abortus 599, as a reference strain, is used for production of diagnostic antigen. In this study we modified FAO/WHO method for production of Brucellae biomass to enhance the productive yield of biomass. The modified media was composed of peptic digest animal tissue (15 g/1), pancreatic digest of casein (15 g/1), yeast extract (10 g/1) and sodium bisulphate (0.10 g/1). The glucose was added during the incubation by fed-batch method (1-30 g/1). The agitation speed and air flow rates were controlled at 300-500 rpm and 4-8 1/min respectively. The culture was harvested after 60 hours. In this method, cell density (packed cell volume) was 9-10% and viable count was 3-3.3xl0u per ml. The biomass production was enhanced to more than 2 times of the FAO/WHO method. The results showed that the media composition and fermentor conditions presented in this study, can be employed to enhance the production of Brucellae biomass for vaccine and antigen preparations in large-scales.
