Brucellosis is a zoonotic disease that causes heavy economic losses and human morbidity. The goal of this study was to prepare a subcellular vaccine for brucellosis. Detoxified Lipopolysaccharide (LPS) and O-polysaccharide (OPS) from B.abortus S99 were conjugated with tetanus toxoid (TT) as a carrier protein. The LPS was extracted by the hot phenol procedure and detoxified. The OPS was extracted using acetic acid. D-LPS-TT and OPS-TT conjugates were prepared by amidation method. The endotoxin activity assay showed 0.125 (EU)/Mg of D-LPS and OPS that were acceptable to use for immunization. The average PS/protein ratios were 0.4 and 0.33 for D-LPS-TT and OPS-TT respectively. The labeling efficiencies were 39 and 44% for D-LPS-TT and OPS-TT respectively. The double immunodiffusion indicated that the conjugates reacted with a single line of identity against the standard Brucella antiserum and tetanus antitoxin. The rabbit thermal induction showed no excess in body temperature with 10Mg of the conjugates injections.
