بررسي خواص ضد جهش زايي و ضد سرطان‌زايي پوست سيب به روش تست ايمز

The study of antimutation and anticarcinogen activity of apple peel by the use of Ames test

سيب‌، منبعي غني از مواد غذايي و تركيباتي مانند فيبر‌، مواد معدني و ويتامين‌ها است. پوست سيب حاوي مقدار قابل توجهي تري ترپنوييد ليپوفيل است كه در لايه كوتيكولي سطح سيب متمركز شده است. هدف از اين پژوهش، تعيين خاصيت ضد جهشي و ضد سرطاني پوست سيب زرد و قرمز در برابر ماده جهش زاي آزيد سديم با استفاده از ميكروزوم و تست ايمز مي‌ باشد. باكتري مورد استفاده سالمونلا تيفي موريوم (100TA‌) است .حضور ماده ضد جهش زايي در پوست سيب در كنار ماده جهش زا، مقدار جهش برگشتي را كاهش مي‌دهد كه در اين بررسي درصد بازدارندگي براي پوست سيب قرمز در حضور 9S 79 درصد و در غياب 9S 4/76 درصد و براي پوست سيب زرد در حضور 9S ‌، 4/69 درصد و در غياب آن 8/62 درصد بود. بازدارندگي بالاي 40 درصد به عنوان ضد جهش قوي شناخته مي‌شود. نتايج اين پژوهش نشان مي‌دهد كه پوست سيب داراي خاصيت ضد جهشي و ضد سرطاني قوي بوده كه اين خاصيت در پوست سيب قرمز بيشتر از پوست سيب زرد است.

Apples are a rich source of nutrient as well as other components like fibr , minerals and vitamins. Apple peel contains considerable amounts of lipophilic triterpenoids which are concentrated in cuticular wax layer and probably have anticancer activity. In this study , we have used acetone extract of red and yellow apple peels. The purpose of this study is to determine antimutation and anticarcinogen effect of red and yellow apple peels against Sodium Azide mutagen substance by the use of Microsome and Ames test. The bacterium used in this test is a strain of salmonella thyphimurium (TA100) that carries a selective mutation in its Histidine Operon. This strain could grow on culture that contains minimum mineral substances and Glucose in the presence of Sodium aside Mutagen substance. presence of anti mutation Ursolic Acid and oleanolic acid along with mutagen substance, reduced quantity of back mutation that the inhibition percentage from mutation was 79% for red apple peel and 69.4 % for yellow apple peel. Inhibition over 40 % recommended as a powerful antimutation substance. Microsome test was used to show the potential ability of antimutation and anticarcinogen activity. Therefore, apple peel has large amount of active Phytochemicals, with Antimutation and Anticarcinogen activity.
