بررسي امكان تعيين ماركر جنسيت در تاسماهي ايراني با استفاده از روش cDNA-AFLP

Investigation of possibility of identification sex marker in persian sturgeon (Acipenserpersicus) by using of cDNA-AFLP technique

ue to high maturation age in sturgeons and lack of morphologic differences between male and female even in brood stocks, sex determination is difficult in these species. Regarding to importance of sex determination of these fish in early stages of life to produce rearing caviar, in this study we used cDNA-AFLP approach in order to analysis of gene expression in 5 female and 5 male Persian sturgeon gonads with using of 63 AFLP primer combinations. Results revealed two cDNA markers in female gonad (TDF1, TDF2) and they verified with RT-PCR in male and female gonads cDNA. But specific primers of TDF1 and TDF2 fragments didnʹt verify in genomic DNA and just in Persian sturgeon female gonad were amplified. According to results of this research and previous researches, it seems that in different sturgeon spices including current studied species, sex chromosomes are not completely differentiated or it is possible that sensitivity of used methods were not sufficient.
