بررسي درون شيشه اي كارايي Acibenzolar-S-methyl بر مهار بيماري سوختگي آتشي در سيب رقم Golden delicious

IN VITRO EVALUATION OF ACIBENZOLAR-S-METHYL ON INHIBITION OF FIRE BLIGHT IN APPLE CV. GOLDEN DELICIOUS

در اين تحقيق، اثر ماده محرك سيستم هاي دفاعي گياه با نام بيون (Acibenzolar-S-methyl, Bion) بر شدت بيماري سوختگي آتشي (آتشك) در سيب رقم Golden delicious بررسي شد. سويه هاي باكتري Erwinia amylovora از نمونه هاي آلوده مناطق قزوين، مشهد، نيشابور و كرج جداسازي و پس از شناسايي، مخلوط چهار سويه به عنوان مايه تلقيح به كار برده شد. گياهچه هاي سيب در محيط MS حاوي BAP و NAA، با بيون، استرپتومايسين و آب مقطر محلول پاشي و سپس به دو روش ته بر كردن ساقه و برش برگ، مايه زني شدند. براساس نتايج، مايه زني با ته بر كردن بلايت شديدتري ايجاد كرد. تيمار بيون بدون بروز اثر مستقيم باكتري كشي، شدت بلايت را در سطح قابل رقابتي با استرپتومايسين كاهش داد. بر اين اساس، به نظر مي رسد ماده محرك بيون بتواند در كنترل بيماري آتشك در سيب رقم گلدن دليشز كه رقم غالب كشور است، موثر باشد.

In this research, the effect of a defense elicitor, Bion ® (Acibenzolar-S-methyl) on fire blight disease in apple cv. Golden delicious was evaluated. Erwinia amylovora strains were isolated from blighted samples collected from Karaj, Qazvine, Mashhad and Neishabur. After identification using biochemical/ physiological tests and PCR, a mixture of four strains was used as inoculum. Golden delicious plantlets were propagated in MS medium supplemented with BAP and NAA and sprayed with Bion, distilled water and streptomycin and inoculated using two different methods. Based on the result, inoculation using shoot cutting method resulted in a higher blight severity than leaf cutting method. Having no bactericidal activity, Bion treatment reduced fire blight severity at a level comparable to streptomycin. Accordingly, it is conculded that Bion elicitor can control fire blight in the most prominent apple cultivar of the country, cv. Golden delicious.
