تعيين ژنوتيپ‌هاي خودسازگار بادام حاصل از يك برنامه اصلاحي و تشخيص آلل‌هايS در برخي ارقام و ژنوتيپ‌هاي خارجي بادام با استفاده از روش PCR

Determination of Self-Compatible Almond Genotypes Obtained from Controlled Crosses and Identification of S-Alleles in some Foreign Cultivars and Genotypes by PCR

يكي از مشكلات توليد بادام مساله خودناسازگاري ارقام بادام است. در اين تحقيق ابتدا تعدادي از ژنوتيپ‌هاي برتر ايراني و خارجي از نظر وضعيت خودسازگاري مورد بررسي قرار گرفتند و در ادامه دورگ‌گيري بين ارقام خودسازگار تونو و سوپر نوا با ده ژنوتيپ مرغوب خودناسازگار انجام شد. وضعيت آلل‌هاي نتاج با استفاده از روش‌هاي كلاسيك (پوشاندن با كيسه) و مولكولي (PCR) مورد بررسي قرار گرفتند. در روش PCR ساده از جفت آغازگر SfF-SfR استفاده شد كه ژنوتيپ‌هاي خودسازگار نواري با طول 450 جفت باز توليد كردند و در ژنوتيپ‌هاي خودناسازگار هيچگونه نواري مشاهده نشد. بر اساس نتايج به دست آمده از تعداد كل 195 دانهال دورگ توليد شده، 110 دانهال خودسازگار و 85 دانهال خودناسازگار تشخيص داده شدند كه نسبت‌هاي به دست آمده براي آلل‌هاي S در جمعيت‌هاي حاصله با نسبت‌هاي مورد انتظار (مندلي) مطابقت داشت و اختلاف معني‌داري بين دو گروه مشاهده نشد. در تحقيق ديگر آلل‌هاي S هر كدام از ارقام و ژنوتيپ‌هاي خارجي مشكوك با استفاده از روش PCR ساده و مركب بررسي شد. در اين تحقيق آلل‌هاي S هر كدام ازارقام و ژنوتيپ‌هاي مورد بررسي مشخص و وضعيت آن‌ها از نظر خود سازگاري و يا خودناسازگاري تعيين شد.

Self-incompatibility is one of the main problems in almond production. In this research work, at the first step, some local cultivars as well as some imported ones were evaluated from the point of their male and female organ compatibility. Then, ten selected commercial self-incompatible cultivars(female parents) were crossed with two self-compatible cultivars(male parents) i. e. Tuono and Supernova. To discriminate self-compatible progenies from others, all seedlings were evaluated through bagging method and molecular marker. With using SfF-SfR primers, self-compatible genotypes produced band of 450bp. However, self-incompatible genotypes did not produce any band. According to the results, out of 195 seedlings, 110 were self-compatible and the other 85 were identified as self-incompatible. This ratio was according to the mendelian law and there was no significant difference between the number of cultivars or genotypes in two groups. In another work, S-alleles of some doubtful cultivars and genotypes as well as their situation from the point of Sf were determined by simple and multiplex PCR.
