شناسايي قارچ هاي همراه با بيماري هاي تنه درختان ميوه هسته دار در استان هاي آذربايجان شرقي و غربي

Identification of the fungal species associated with trunk diseases of stone fruit trees in East and West Azerbaijan provinces

بيماري هاي تنه به عنوان يك تهديد بالقوه براي توليد و كشت و پرورش درختان ميوه هسته دار در دنيا مطرح مي باشند. عوامل زنده مختلفي در ايجاد اين بيماري ها دخالت دارند كه در بين آن ها عوامل قارچي، سهم قابل توجهي را به خود اختصاص مي دهند. علي رغم شيوع بيماري هاي تنه در باغ هاي درختان ميوه هسته دار استان هاي آذربايجان شرقي و غربي، عوامل قارچي همراه با اين بيماري ها ناشناخته باقي مانده اند. در اين بررسي گروه هاي قارچي همراه با بيماري-هاي تنه درختان ميوه هسته دار در استان هاي آذربايجان شرقي و غربي بر اساس داده هاي ريخت شناختي و داده هاي توالي نوكليوتيدي ناحيه rDNA-ITS مورد شناسايي قرار گرفت. از مجموع 128 نمونه تعداد 101 جدايه قارچي جداسازي و خالص سازي گرديد. گونه هايchrysosperma Cytospora با فراواني 76/23 درصد، Alternaria alternata با فراواني 86/13 درصد، Paecilomyces variotii با فراواني 9/9 درصد و Fusarium proliferatum با فراواني 91/8 درصد به عنوان گونه هاي غالب شناسايي شدند. در مورد جدايه هاي متعلق به جنس آلترناريا هويت گونه با استفاده از داده هاي توالي ژنrpb2 مورد تاييد قرار گرفت. هم چنين گونه هاي Epicoccum nigrum، Cladosporium cladosporioides، Phoma glomerata، Paraconiothyrium africanum، Sarocladium strictum، Chaetomium globosum، Fusarium acuminatum، F. tricinctum، Cytospora cincta، C. pruinosa، Botryosphaeria sarmentorum، Phaeoacremonium aleophilum با درصد فراواني كمتر جداسازي شدند. بين گونه هاي جداسازي شده Cytospora chrysosperma، C. cincta، C. pruinosa، sarmentorum B.، P. aleophilum وP. africanum به عنوان بيمارگر هاي شناخته شده روي ميزبان هاي چوبي به شمار مي-روند. با شناسايي گونه هاي قارچي همراه با بيماري هاي تنه درختان ميوه هسته دار، اعمال راه كارهاي مناسب براي مديريت بيماري در آينده امكان پذير خواهد شد.

Trunk diseases are considered as a potential threat for the cultivation and production of stone fruit trees all over the world. Various biotic agents are involved in trunk diseases, of those fungal gropus are the most common ones playing prominent role in these diseases. Given the incidence of the trunk diseases on stone fruit trees in the orchards of East and West Azerbaijan provinces, the identity of fungal species associated with disease symptoms mainly remain unknown. In the present study the fungal species associated with the trunk diseases symptoms on stone fruit trees in this region were characterized using morphological and molecular data. A total number of 101 fungal isolates were isolated from 128 diseased samples. Our results revealed that Cytospora chrysosperma with the isolation freqeuncey on 23.76 % represented the most dominant species followed by Alternaria alternata (13.86 %), Paecilomyces variotti (9.9 %) and Fusarium proliferatum (8.91 %). The identity of Alternaria isolates was confirmed using seqeunce data of rpb2 gene. The other fungal species which were isolated with lower frequencies included Epicoccum nigrum, Cladosporium cladosporioides, Phoma glomerata, Paraconiothyrium africanum, Sarocladium strictum, Chaetomium globosum, F. acuminatum, F. tricinctum. C. cincta, C. pruinosa, Botryosphaeria sarmentorum and Phaeoacremonium aleophilum. Among the fungal species isolated in this study Cytospora chrysosperma, C. cincta, C. pruinosa B. sarmentorum, P. aleophilum and P. africanum have been reported as the causal agents of trunk diseases on stone fruit trees. With the identification of fungal species associated with trunk diseases on stone fruit trees, it will be possible to practice suitable management strategies for these diseases.
