بررسي اثر ضد باكتري مخلوط سيلر AH26 با دو نوع آنتي‌بيوتيك متفاوت بر انتروكوكوس فكاليس به‌روش آزمايشگاهي

In vitro evaluation of the antibacterial effects of AH26 combined with two different antibiotics on Enterococcus faecalis

چكيده مقدمه: براي جلوگيري از رشد باكتري‌هاي باقي‌مانده پيشنهاد شده كه مواد پركننده كانال و سيلرها خصوصيت ضد ميكروبي داشته باشند. هدف از اين مطالعه بررسي اثر ضد باكتري مخلوط AH26 با دو نوع آنتي‌بيوتيك متفاوت بر انتروكوكوس فكاليس بود. مواد و روش‌ها: در اين مطالعه آزمايشگاهي، خاصيت ضد ميكروبي آموكسي‌سيلين و مينوسيكلين كه به‌‌صورت جداگانه به سيلر AH26 اضافه شده بودند بر روي انتروكوكوس فكاليس مورد بررسي قرار گرفتند. در گروه اول سيلر AH26 با آنتي‌بيوتيك آموكسي‌سيلين 25 درصد، گروه دوم سيلر AH26 با آنتي‌بيوتيك مينو‌سيكلين 25 درصد، در گروه سوم سيلر AH26 و در گروه چهارم آب مقطر استفاده شد. نمونه‌ها در محيط كشت آگار كه با انتروكوكوس فكاليس تلقيح شده بود قرار گرفتند. نمونه‌ها به مدت هفت روز در دماي 37 درجه سانتي‌گراد تحت شرايط بي‌هوازي انكوبه شده و قطر مهار رشد در24، 48، 72 ساعت و يك هفته پس از آن بر حسب ميلي‌متر اندازه‌گيري شد. داده‌ها با آزمون‌هاي آماري واريانس دو سويه، Repeated measures ANOVA و Tukey HSD مورد ارزيابي قرار گرفتند (05/0 = ?). يافته‌ها: نتايج نشان داد افزودن هر دو نوع آنتي‌بيوتيك با غلظت 25 درصد به سيلر AH26، نواحي ممانعت از رشد وسيعي نسبت به سيلر خالص ايجاد كرد (001/0 > p value). در گروه 1 و 2 با افزايش دوره‌ي انكوباسيون از 24 ساعت به 72 ساعت، تفاوت معني‌داري در نواحي ممانعت از رشد باكتري ايجاد شد، اما تفاوت بين 72 ساعت و يك هفته معني‌دار نبود. بيش‌ترين خاصيت ضد ميكروبي متعلق به گروه حاوي آموكسي‌سيلين بود. نتيجه‌‌گيري: مخلوط كردن سيلر AH26 با آموكسي‌سيلين و مينوسيكلين خاصيت ضد ميكروبي سيلر در برابر انتروكوكوس فوكاليس را افزايش مي‌دهد. كليد واژه‌ها: آموكسي‌سيلين، انتروكوكوس فوكاليس، سيلر AH26، مينوسيكلين

Abstract Introduction: To prevent growth of residual bacteria, it has been proposed that sealers and filling materials should have antibacterial properties. The aim of this study was to evaluate the antibacterial effects of AH26 sealer combined with two different antibiotics on Enterococcus faecalis. Materials and methods: In this in vitro study, the antimicrobial effects of amoxicillin and minocycline which were added separately to AH26 sealer were evaluated on Enterococcus faecalis. In group 1, 25% amoxicillin with AH26 sealer, in group 2, 25% minocycline with AH26 sealer, in group 3, pure AH26 sealer, and in group 4, distilled water were used. The samples were placed in agar plates inoculated with Enterococcus faecalis. All the plates were incubated for 7 days at 37°C under anaerobic condition, and zones of growth inhibition were measured in millimeters at 24-, 48-, 72- and 7-day intervals. Data were analyzed using 2-way repeated measures ANOVA and multiple comparisons were made by Tukey tests (?=0.05). Results: The results showed that adding antibiotics at 25% concentrations to AH26 sealer created significantly larger zones of growth inhibition compared to the sealer alone (p value < 0.05). In groups 1 and 2, there were significant differences in zones of growth inhibition with an increase in incubation period from 14 to 72 hours; however, there were no significant differences between 72-hour and 1-week intervals. The maximum antibacterial activity was observed in the amoxicillin group. Conclusion: Mixing AH26 sealer with amoxicillin and minocycline significantly increases the antibacterial property of the sealer against Enterococcus faecalis.
