Evaluation of sperm quality, maturation and DNA integrity in adult mice treated with sulpiride

800x600 Normal 0 false false false EN-US X-NONE AR-SA MicrosoftInternetExplorer4 /* Style Definitions */ table.MsoNormalTable {mso-style-name:"Table Normal"; mso-tstyle-rowband-size:0; mso-tstyle-colband-size:0; mso-style-noshow:yes; mso-style-priority:99; mso-style-parent:""; mso-padding-alt:0in 5.4pt 0in 5.4pt; mso-para-margin:0in; mso-para-margin-bottom:.0001pt; mso-pagination:widow-orphan; font-size:10.0pt; font-family:"Times New Roman","serif";} زمينه و هدف: مصرف بعضی از داروهای آنتی‌سايكوتيك تاثير شديدی بر باروری جنس نر دارد. اثرات هيپوفيزی و تغيير در غلظت سرمی بعضی از هورمون‌ها مثل پرولاكتين، LH و FSH تاثيرات مختلفی بر توليد اسپرم دارد. در اين مطالعه اثر داروی سولپرايد بر روی كيفيت، ميزان آسيب DNA و وضعيت بلوغ هسته اسپرم مطالعه شد. روش بررسی: 24 موش نر سوری بالغ (سن: 8-6 هفته) به سه گروه تيمار، كنترل شم و كنترل تقسيم شدند. به‌مدت 45 روز روزانه به گروه تيمار محلول سولپرايد به‌ميزان mg/kg40 و به كنترل شم حلاّل دارو به‌صورت داخل صفاقی تزريق شد. گروه كنترل چيزی دريافت نكرد. پس از اين مدت تمامی موش‌ها به روش جابه‌جايی گردن كشته شده، اپيديديم به روش جراحی خارج شد و درml 1 محيط كشت Human Tubal Fluid (HTF) به‌مدت نيم ساعت در داخل انكوباتورCO2  به منظور شناورسازی اسپرم‌ها گذاشته شد. تعداد، قدرت تحرك و قدرت زيست‌پذيری اسپرم‌ها مورد بررسی قرار گرفت. هم‌چنين كيفيت بلوغ هسته و ميزان آسيب DNA اسپرم توسط رنگ‌آميزی آنيلين بلو و آكريدين اورنج ارزيابی شد. يافته‌ها: نتايج نشان داد در موش‌هاي گروه درمانی، كاهش چشمگيری در تعداد و قدرت تحرك اسپرم‌ها مشاهده شد و تعداد اسپرم‌هاي غيرطبيعی در مقايسه با دو گروه ديگر افزايش معنی‌داری داشت (05/0P<). كاهش مشخصی در قدرت زيست‌پذيري و بلوغ هسته اسپرم‌ها ديده شد و ميزان آسيب DNA در مقايسه با گروه‌های ديگر افزايش چشمگيری يافت (05/0P<).  نتيجه‌گيری: مشخص شده داروی آنتی‌سايكوتيك سولپرايد دارای اثرات منفي بر پارامترهای اسپرم در موش‌های درمان شده است و در تعدادی از نمونه‌ها هيپوگناديسم ثانويه ايجاد می‌كند.

Background: Use of certain antipsychotic drugs has severe effects on fertility in males. Hypothalamus and hypophysial impressions and changes in plasma hormones concentration like prolactin, LH and FSH can affect sperm production. In this study, we investigated the effects of sulpiride on sperm quality, maturation and DNA damage. Methods: Twenty for adult male mice (age: 6-8 weeks) were divided into three groups. The treatment group received 40 mg/kg sulpiride solution and the control sham group was given carrier of the drug intraperitoneally (IP) daily for 45 days but the control group received nothing. Finally, all the mice were sacrificed by cervical dislocation and their cauda epididymis were removed surgically. The excised specimens were placed in 1 ml HTF medium and incubated for 30 min in CO2 incubator to allow the spermatozoa to swim out. Later, sperm count, motility and viability were analyzed. Additionally, sperm chromatin quality and DNA integrity were assessed by aniline blue and acridine orange staining. Results: Significant decrease in sperm motility and count were observed in the treatment group while the number of abnormal sperm increased as compared with the other two groups. Sperm viability and DNA maturation showed significant reduction and the rate of DNA damage increased in comparison with the control sham and the control groups (P<0.05). Conclusion: The study showed that sulpiride has negative effects on sperm parameters in treated animals and in some cases it could cause secondary infertility.
