ارتباط چندشكلي نواحي 5`UTR و اگزون يك ژن ميوستاتين با صفت دوقلوزايي در نژاد بز مرخز

Association of Polymorphism in 5`UTR and Exon1 Regions of Myostatin Gene with Twining Trait in Markhoz Goat Breed

در اين مطالعه هدف بررسي چندشكلي هاي ژن ميوستاتين و ارتباط آنها با صفت دوقلوزايي در150 راس نژاد بز مرخز ايران بود. قطعه 573 جفت بازي از ژن ميوستاتين حاوي يك حذف شدگي 5 نوكليوتيدي(TTTTA/ 206) و يك جهش تك نوكليوتيدي ( A/T339) به ترتيب در ناحيه 5`UTR و اگزون يك تكثير شد. هضم محصولات PCR با استفاده از آنزيم محدودگر DraIو HinIII به صورت جداگانه براي هر جايگاه انجام گرفت. كه نتايج آن حاكي از دو ژنوتيپ AA و AB در ناحيه 5`UTR و سه نوع ژنوتيپ AT , TT و AA در اگزون يك بود. برآورد نسبت احتمالات (OR) دوقلوزايي در شكم زايش اول نسبت به دوم و سوم به ترتيب برابر با 4/1 (05/0 < P)، 4 (1/0 > P) و در شكم زايش دوم نسبت به سوم نيز60/4 (05/0 > P) بود. مقايسه ژنوتيپي ژن ميوستاتين نشان داد كه ژنوتيپ AA نسبت به ژنوتيپ TT ( 63/13= كاي اسكور، 84/1= OR) و ژنوتيپ TT نسبت به ژنوتيپ AT با( 46/8 = كاي اسكور، 28/0= OR) در سطح احتمالي01/0 داراي اختلاف معني دار هستند. برآورد نسبت احتمالات ژنوتيپ AB نسبت به AA در ناحيه 5`UTR برابر با 07/0 (1/0 > P) بود. هم چنين نتايج اين تحقيق رايطه معني داري صفت دوقلوزايي با ژن ميوستاتين را در بزهاي مرخز تاييد نموده و مطالعه بيشتر روي نژادهاي بومي ديگر ايران را پيشنهاد مي كند.

This study aimed to investigate the myostatin gene polymorphisms and their relationship with the twining trait in 150 Iranian Markhoz goats. A 573 bp fragment of the myostatin gene was amplified, which contained a deletion 5 bp indel (206 TTTTA/) and a single nucleotide polymorphism substitution (339 T/A) in 5`UTR and exon1 regions, respectively. The PCR products were separately digested for each position using DraI or HinIII restriction enzyme endonuclease. The digestion results revealed two AA and AB genotypes in the 5`UTR region and three AA, AT, and TT genotypes in exon1 region of myostatin gene. Odds Ratio (OR) statistics for twinning trait was estimated 1.4 (P > 0.05) and 4 (P < 0.1) in the first parity compared to the second and third parity, respectively. Also, the odds ratio was 4.60 (P < 0.05) when the second parity was compared to the third parity. Genotype comparison of the myostatin gene showed that the AA genotype showed a significant difference with the TT genotype (chi square=13.63, OR=1.84) and the TT genotype showed a significant difference with the AT genotype (chi-square=8.46, OR= 0.28) (P=0.01). The OR of the twining rate was estimated 0.07 (P < 0.1) for the AB genotype compared to the AA genotype in 5`UTR region. The results of this research confirm a significant association between the myostatin gene and the twining rate in Markhoz goats and suggest more investigation in this field on other Iranian native goat breeds.
