ارزيابي خواص ضدالتهابي مشتقات پارا- متيل سولفونيل- 3 و 4-دي آريل -4- تيازولين-2- تيون‌ها به روش كاراژينان در موش سوري

Anti-inflammatory Properties of 4-(4-Methylsulfonylphenyl)-3-phenyl-2(3H)-thiazole thione Derivatives by Carrageenan Method in Mice

سابقه و هدف: آنزيم سيكلو اكسيژناز 2 از مهم‌ترين آنزيم‌های مسير سنتز پروستاگلاندين‌ها و پيدايش التهاب در بدن انسان است. در پژوهش حاضر اثر هشت تركيب جديد سنتز شده با پتانسيل مهار سيكلواكسيژناز 2 به روش ادم كف پايی ناشی از كاراژينان در موش سوری مورد ارزيابی قرار گرفت. مواد و روش‌ها: دوزهای مختلف تركيبات (10، 25 و 50 ميلی‌گرم بر كيلوگرم)، سلكوكسيب (كنترل مثبت، 10 ميلی‌گرم بر كيلوگرم) و نرمال سالين- تويين (كنترل منفی) به روش داخل صفاقی به موش‌ها (جنس ماده، وزن 30-25 گرم، تعداد در هر گروه 6 سر) استعمال شدند. بعد از نيم ساعت، كاراژينان به شكل زير جلدی به بافت كف پای چپ حيوان در تمامی گروه‌ها تزريق شد. حجم پا در زمان‌های 1، 3 و 5 ساعت بعد از تزريق اندازه‌گيری و درصد تغييرات حجم پا به نسبت ساعت صفر اندازه‌گيری شد. نتايج به صورت ميانگين± انحراف معيار گزارش شدند. p<0.05 به عنوان حداقل سطح معنی‌داری در نظر گرفته شد. يافته‌ها: در دوز ده ميلی‌گرم بر كيلوگرم تمامی تركيبات اثر معنی‌دار ضدالتهابی را فقط در ساعت اول نشان دادند. در دوز 25 ميلی‌گرم بر كيلوگرم، برخی تركيبات در تمامی زمان‌های مورد بررسی اثر ضدالتهابی معنی‌دار در مقايسه با گروه كنترل داشتند. در دوز پنجاه ميلی‌گرم بر كيلوگرم، تمامی تركيبات در همه زمان‌های يك، سه و پنج ساعت در مقايسه با گروه كنترل نرمال سالين- تويين اثر ضدالتهابی معنی‌دار نشان دادند. استنتاج: اين مطالعه نشان داد، مشتقات پارا- متيل سولفونيل- 3 و 4-دی آريل -4- تيازولين-2- تيون‌ها می‌توانند فعاليت ضد التهابی مطلوبی از خود نشان دهند.

Background and purpose: Cyclooxygenase II enzyme is one of the most important enzymes of prostaglandin synthesis pathway that also causes inflammations in human body. In the present study, the anti-inflammatory activity of eight new synthesized compounds with COX II inhibitory potential was evaluated in carrageenan induced paw edema in mice. Material and methods: Different doses of the compounds (10, 25 and 50 mg/kg), celecoxib (positive control, 10 mg/kg) and normal saline-tween (negative control) were intraperitoneally administered to the mice (female, 25-30 g, n=6). After 30 min, carrageenan was injected subcutaneously into the subplantar tissue of the left paw in all groups. The volume of paw was measured in different treatment groups at 0, 1, 3 and 5 hours following the carrageenan injection and percentage changes of paw volume was calculated. Data were reported as Mean ± SD. T-test was used to compare the results and P < 0.05 was considered significant. Results: At the 10 mg/kg dose, all compounds showed significant anti-inflammatory activity just in 1st hour. At the 25 mg/kg dose, some compounds had significant anti-inflammatory activity in all investigated times, and at 50 mg/kg dose, all compounds showed significant anti-inflammatory activity in 1st , 3rd and 5th hour compared to the group injected with normal saline- tween. Conclusion: This study found 4-(4-Methylsulfonylphenyl)-3-phenyl-2(3H)-thiazole thione derivatives with good anti-inflammatory activities.
