بررسي عصاره الكلي برگ گياه حنا بر روي رگ زايي پرده كوريوآلانتوئيك و برخي ناهنجاري هاي مورفولوژيك در جنين جوجه

The effects of henna leaf alcoholic extract on angiogenesis of chorioallantoic membrane and some embryonic morphological abnormalities of chick embryos

زمينه و هدف: آنژيوژنز، تشكيل مويرگ های جديد از عروق اوليه، در فرآيند های فيزيولوژيك مانند توليد مثل و بهبود زخم و پاتولوژيك مانند ديابت و تومور نقش دارد. رشد و توسعه تومور با رگ زايی ارتباط دارد. اين تحقيق با هدف بررسی اثرات عصاره الكلی برگ حنا (Lawsonia inermis) بر آنژيوژنز پرده كوريوآلانتوئيك و ناهنجاری های مورفولوژيك جنين جوجه انجام شده است. روش بررسی: در اين پژوهش تجربی، 40 عدد تخم مرغ نطفه دار نژاد Hy-line به طور تصادفی در 4 گروه: شاهد، شاهد آزمايشگاهی، گروه تجربی 1 و گروه تجربی 2 تقسيم شدند (10=n). روز دوم انكوباسيون در شرايط استريل، در تخم مرغ ها پنجره ايجاد گرديد. روز هشتم پرده كوريوآلانتوئيك در گروه شاهد آزمايشگاهی با نرمال سالين و در گروه تجربی 1 و 2 با عصاره الكلی برگ حنا به ترتيب با دوزهای 50 و mg/kg200 تيمار گرديد. روز دوازدهم از عروق ناحيه تيمار عكس برداری و تعداد و طول انشعابات عروقی اندازه گيری شد. در جنين های برداشت شده، وزن و طول سری- دمی (CR) و ناهنجاری های مورفولوژيكی مورد بررسی قرار گرفت. يافته ها: مقايسه ميانگين تعداد و طول انشعابات عروقی، وزن و طول سری- دمی جنين جوجه در گروه شاهد با شاهد آزمايشگاهی اختلاف معنی دار نشان نداد (05/0>P). در گروه تجربی 2 ميانگين تعداد و طول انشعابات عروقی و در گروه تجربی 1 ميانگين طول انشعابات عروقی و طول سری- دمی نسبت به گروه شاهد كاهش معنی داری نشان داد (05/0>P). ميانگين متغيرها در گروه های تجربی اول و دوم اختلاف معنی دار نشان نداد (05/0>P). در بررسی مورفولوژيكی جنين ها هيچگونه ناهنجاری مشاهده نشد. نتيجه گيری: نتايج اين تحقيق نشانگر اثرات مهاركننده رگ زايی عصاره الكلی برگ حنا بر آنژيوژنز جنين جوجه است؛ اما بر خصوصيات مورفولوژيكی جنين جوجه تأثير قابل توجهی ندارد.

Background and aims: Angiogenesis, formation of new vessels from the primary vessels is involved in physiological condition such as reproduction, wound healing and pathological like diabetes and tumor. Tumor progression associates with persistent unregulated angiogenesis. This study aimed to investigate the effect of alcoholic extract in henna (Lawsonia inermis) leaf on the angiogenesis of chorioallantoic membrane and morphological abnormalities of chick embryos. Methods: In this experimental study, 40 fertilized eggs of Hy-line race were randomly divided into 4 groups: control group, sham-exposed group and two empirical groups (n=10). In the second day of incubation in sterile condition, a window was opened on eggs. On 8th day, chorioallantoic membrane (CAM) was treated at sham-exposed group with normal saline and at empirical groups with alcoholic extract in henna (Lawsonia inermis) leafs (50, 200 mg/kg). On 12th day CAMs, the length and weight of vessels were examined and morphological disorders were studied. Data were calculated and analyzed statistically using t-test and ANOVA. Results: Weight and length CR of chick embryo at the control group with the sham-exposed group showed no significant difference in the mean comparison of number and length of vessels. The mean of number and length in the second empirical group and in the first empirical group at the comparison of vesselʹs length and the CR length than the control group showed significant decrease (P<0/05). Variables mean in two empirical groups showed no significant difference. Morphological abnormalities were not observed in fetuses. Conclusion: According to the results of this research, alcoholic extract of henna leaf inhibits angiogenesis on chorioallantoic membrane, but it had no significant effect on chick embryoʹs morphological characteristics.
