بررسي قابليت چسبندگي باكتري پروبيوتيكي لاكتوباسيلوس كازيي به لايه سلولي Hep2 cell

Evaluation of adherence of probiotic bacteria Lactobacilus casei to Hep2 cell line

اولين قدم براي انتخاب يك سويه به عنوان پروبيوتيك تعيين قابليت چسبندگي آن سويه به مخاط ميزبان مي باشد تا علاوه بر پيشگيري از چسبندگي باكتري هاي بيماري زا، با ترشح برخي مواد، محيط را براي رشد آنها نامساعد سازد. اين مطالعه با هدف ارزيابي قابليت چسبندگي سويه پروبيوتيكي لاكتوباسيلوس كازيي به سلول هپتو انجام شد. در ابتدا لايه سلولي هپتو به پليت 6 چاهك منتقل و در محيط كشت RPMI حاوي سرم جنين گوساله و آنتي بيوتيك رشد داده شد. سپس باكتري پروبيوتيكي كشت شده در محيط MRS براث، به ميزان مشخص به هپتوسل اضافه شد. پس از تعيين زمان مطلوب براي تماس باكتري با لايه سلولي، هپتوسل شستشو داده شد. سلول ها رنگ آميزي و با روش هاي بافت شناسي تثبيت شدند. قابليت يا عدم قابليت و طرح چسبندگي باكتري پروبيوتيكي در زير ميكروسكوپ نوري مشاهده گرديد. نتايج نشان داد كه زمان 3 ساعت زمان مناسب براي آزمايش بوده و لاكتوباسيلوس كازيي قابليت چسبندگي به سلول مخاط دهاني- حلقي را دارا مي باشد. با توجه به نتايج به دست آمده، استفاده از باكتري لاكتوباسيلوس كازيي به عنوان پروبيوتيك در محصولات بهداشتي مربوط به دهان و دندان پيشنهاد مي گردد.

The first step to choose a strain as probiotic is capability for mucosal adhesion. The adherence is a preliminary factor to prevent pathogenic bacteria, with the secretion of some substances, as well as inhibiting the growth and colonization of them. Hep-2 cell, from pharynx mucosa carcinoma, is so far used to study the adhesion of E. coli bacteria. Therefore, in the present study, Hep-2 cell layer will be used to survey the adhesion capability of probiotic bacteria to oral epithelial cells. In this study, the adhesion of the probiotic strain Lactobacillus casei to Hep-2 cells was examine The Hep-2 cells were grown in RPMI medium containing fetal calf serum and antibiotics and incubated appropriate atmosphere. After 24 hours, cells were washed and new medium without antibiotics were added. The probiotic bacteria cultured in MRS broth added to the Hep-2 cell with certain rate and incubated either 1, 2 or 3 hours to determine the appropriate time. Then the cells were stained and fixed with histological methods. Adhesion ability and design of probiotic bacteria, was observed under optical microscope. The present study showed that the tested strain was able to adhere to oro- pharyngeal cell line. As well, the optimum time to test the ability was 3 hours. These results indicated that the probiotic bacterium Lactobacillus casei is suitable for application in health products. For the further investigation, animal and human tests are necessary.
