بررسي تغييرات بافت‌شناسي پيش معده مرغان گوشتي راس در سنين مختلف رشد

An investigation on histological changes of the Ross broiler’s proventriculus at different ages

    با توجه به نقش پیش معده پرندگان در هضم وگوارش مواد غذایی و نیز وجود تفاوت‌های مورفولوژیكی و عملكردی زیاد در بین گونه‌های مختلف پرندگان، این مطالعه به انجام رسید. در این مطالعه 54 قطعه جوجه گوشتی راس نر و ماده در سنین 3 روزگی، 7 روزگی و پس از آن به ‌فاصله یك هفته تا سن 8 هفتگی مورد استفاده قرار گرفت. پیش معده جدا و در محلول فرمالین بافر 15 درصد تثبیت شد. برش‌های بافتی 5 میكرونی از نمونه‌ها تهیه و با هماتوكسیلین ائوزین و رنگ‌آمیزی‌های اختصاصی آلسین بلو، اورسئین، ماسون تریكروم، پاس و گوموری مخصوص رشته‌های رتیكولر رنگ‌آمیزی شدند. نتایج نشان داد كه با افزایش سن، ارتفاع و آناستوموز چین‌های مخاطی، تجمعات لنفاوی، تعداد و فراونی غدد سطحی مخاطی، تعداد و اندازه غدد عمقی مخاطی، ضخامت مخاط، لایه ماهیچه مخاطی، طبقه ماهیچه‌ای بویژه لایه حلقوی داخلی، طبقه سروزی و بافت چربی آن در پیش معده افزایش یافته است. انشعابات مجاری غدد عمقی نیز كه به اشكال مختلف با غالبیت چند وجهی، بیضی و مثلثی مشاهده شدند، با افزایش سن بیشتر شد. تفاوت بافتی قابل توجهی بین جنس‌های نر و ماده مشاهده نشد. در مجموع می‌توان نتیجه گرفت كه ساختار بافت شناسی پیش معده در مرغان گوشتی راس با مرغان بومی دارای تفاوت‌های زیر است: ماهیچه مخاطی پیوسته و ضخیم بود، غدد عمقی از نوع مركب لوله‌ای و در بافت پیوندی پارین قرار گرفته‌اند، تجمعات لنفاوی در زیر و داخل غدد عمقی و نیز در زیر مجرای مركزی غدد حضور داشتند.

As the proventriculus plays an important role in food digestion and there are differences in morphological and functional characteristics among different avian species, this study was carried out. A total of 54 female and males Ross broiler chicks at age 3, 7 days and then at weekly intervals until week 8 were used. The proventriculus were collected and fixed in 15% buffered formalin solution. Histologic sections of 5µm of thickness were obtained and they were stained routinely with hematoxylin-eosin and special stainings: Alcian blue, Orcein, Masson’s trichrome, PAS and Gomori’s method for reticular fibers. The results showed that by age increasing, the height of mucosal folds (particularly anastomoses), lymphatic aggregations, the number and frequency of superficial mucosal glands, the size and number of deep mucosal glands, thickness of muscularis mucosa, tunica muscularis (particularly inner circular) and serosa (particularly adipose tissue) were also increased. The shape of the deep glands was varied but polygonal, elliptical and triangular structures more prominent. The duct branching of these glands were increased with age. There was not a significant histological difference between male and female chickens. Our observations on the general histological structures of proventriculus were similar to native chickens stated in literature, but there were also some differences. The muscularis mucosa is thick and continuous. The deep glands are compound tubular and located in the lamina propria. Lymphatic aggregations are present within the connective tissue between the compound glands, and beneath their central unique ducts.
